<p>PURPOSE: To obtain the subject new antibody, capable of binding to a 7S domain of a pepsin hydrolyzate of a human IV type collagen without binding to other domains, human I, III and V type collagens and useful as a hepatic fibrotic marker for clinical diagnosis, etc. CONSTITUTION: This monoclonal antibody against a human IV type collagen is produced by the hydrolysis of a human IV type collagen with pepsin and has the properties of binding to a 7S domain of a human IV type collagen without binding to other domains, human I, III and V type collagens, having about 51000 molecular weight of a heavy (H) chain and about 28000 molecular weight of a light (L) chain when measured according to a reduced form sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis, an IgG1κtype, capable of accurately measuring the human IV type collagen in blood serum for a short time and useful as a hepatic fibrotic marker for clinical diagnosis, etc. The antibody is obtained by fusing a murine lymphocyte sensitized with the human IV type collagen to a cell of a myeloma, then cloning the fused cell and culturing the cloned fused cell.</p> |
