PURPOSE: To change the unsaturation degree of a fatty acid such as rice plant or corn and carry out the functional modification by integrating a reconstructed gene containing a cDNA, capable of coding a specific amino acid sequence and connected to a promoter and a terminator into a gramineous plant. CONSTITUTION: The unsaturation degree of a fatty acid of a plant is modified by integrating a reconstructed gene prepared by connecting a cDNA of a Δ9-fatty acid desaturase capable of coding an amino acid sequence of the formula, especially a cDNA of a base sequence of the formula to a promoter and a terminator in the sense and anti-sense directions into a gramineous plant. This cDNA is obtained by amplifying a corresponding DNA fragment of a rice plant according to polymerase chain reaction(PCR) using an oligomer synthesized from the amino acid sequence of a Δ9-stearoyl-acyl carrier protein(ACP) desaturase of a dicotyledonous plant in anticipation of a base sequence, screening a cDNA library with the fragment, localizing the longest clone of the resultant positive clone in a chloroplast of the rice plant and capable of coding the Δ9-stearoyl-ACP desaturase. |
