PURPOSE: To obtain the subject new gene capable of coding a phytoene dehydrogenase derived from a photosynthetic bacterium of the genus Erythrobacter and economically producing a natural product β-carotene, useful as a colorant for foods, a functional food, etc., and having high safety. CONSTITUTION: This new gene is capable of coding a phytoene dehydrogenase derived from a photosynthetic bacterium of the genus Erythrobacter and economically producing a natural product β-carotene useful as a colorant for foods, a functional food having cancer preventing effects, etc. Furthermore, the gene is obtained by culturing a photosynthetic bacterium of the genus Erythrobacter [e.g. Erythrobacter longus Och101 (ATCC14126)] in a culture medium, proliferating the bacterium, then collecting the bacterium, suspending the collected bacterium in a buffer solution, adding a proteinase thereto, incubating the proteinase for 1hr, carrying out the extraction with phenol/chloroform, recover the total DNA, dissolving the total DNA in a buffer solution, treating and purifying the resultant DNA with an RNase, then treating the purified DNA with a restriction enzyme, preparing a genome library, screening the library with a probe, sorting out a positive clone, recovering the DNA and treating the recovered DNA with a restriction enzyme. |
