PURPOSE: To obtain a new plasmid vector, comprising a specific DNA fragment and a DNA fragment containing a kanamycin-resistant gene capable of functioning in a bacterium of the genus Thermus and suitable for breeding a highly thermophilic bacterium of the genus Thermus and expressing a useful gene using the bacterium as a host at a high efficiency. CONSTITUTION: This new plasmid vector comprises a DNA fragment containing a replication proliferation region of a cryptic plasmid such as pNHK101 held by Thermus spp. TK 10 strain (FERM P-6750) derived from a bacterium of the genus Thermus and a DNA fragment containing a kanamycin-resistant gene capable of functioning in the bacterium of the genus Thermus and has a large number of copies. |
