<p>PURPOSE: To obtain new cephem compound useful as an antimicrobial agent, showing antibacterial action on gram-positive and gram-negative bacteria, by making a specific cephalosporin derivative containing a halomethyl group into a phosphonium compound and reacting the compound with an aldehyde to introduce a double bond. CONSTITUTION: A compound of formula I [R1 is a (protected) amino group; R<2> a group of formula II (R<5> is H, an OH-protecting group or an aralkyl; R<6> is H, an amino-protecting group or an aralkyl); R<3> is H, a metal atom, a carboxyl group-protecting group, an ester residue capable of forming an ester hydrolyzable in vivo or a negative charge; X is a halogen) is reacted with a phosphine of the formula P(R<10> )3 (R<10> is butyl or phenyl) to give a phosphonium salt compound corresponding to the compound of formula I. The compound is reacted with an aldehyde compound of the formula X-CH2 -CHO and the reaction product is further reacted with a thiazole compound to give a new cephem compound (salt) of formula III (R<4> is thiazolium or pyrimidinium).</p> |
