PURPOSE: To examine the mutual compensation of enzyme with high sensitivity for quantitative analysis of high-molecular-weight and low-molecular-weight examinee by comparing the invert ratio of matrix with the invert ratio of matrix obtained by using examinee of known concentration. CONSTITUTION: Matrix reactive with specimen, enzyme donor polypeptide coupler, examinee coupling protein, enzyme acceptor polypeptide and active enzyme complex is combined in medium to form reactive mixture. The invert ratio of the matrix in the reactive mixture is measured and compared with the invert ratio of matrix obtained by using examinee of known concentration. The amount of examinee in specimen is thus measured. The enzyme donor is blended protein which has aα-donor domain, where active enzyme complex can be formed by interaction with enzyme acceptor, and a protein domain containing protein antigen or antigen epitope. Enzyme donor polypoptide which is prepared by recombination DNA technology is used. |
