PURPOSE:To provide a method for counting leucocytes classified at least in five groups by using a reagent for measuring the leucocyte in four grouping and a reagent for measuring basophilic corpuscles. CONSTITUTION:A reagent for measuring the leucocyte in four grouping is prepared from a sufficient quantity of ionic surface active agent which admits dissolution of red corpuscles and gives a damage to the cellular membrane of a leucocyte, an organic compound having a sufficient quantity of anionic radicals coupling with the cation component of a leucocyte to give a form difference between leucocytes, a nonionic surface active agent in a sufficient quantity to make soluble the undissolved portion due to coupling of the cellular component with the ionic surface active agent, and a buffer material. This reagent is added to part of an all-blood specimen, and the cell size/form information is acquired, and the corpuscles are counted upon being classified in at least four groups, i.e., three groups of lymphocytes, mono-corpuscles, and acidphilic corpuscles and one group of neutral-philic corpuscles and basophilic corpuscles, Another reagent for measuring basophilic corpuscles is added to other part of the all-blood specimen, and at least the cell size information is acquired, and the basophilic corpscles are counted upon being classified. The luecocytes are classified into five groups on the basis of the counts. |
