<p>PURPOSE:To obtain a heparan sulfate 6-O-sulfate group transferase capable of selectively introducing sulfate group to the 6-site of glucosamine of heparin and heparan sulfate. CONSTITUTION:This heparan sulfate 6-O-sulfate group transferase having the following properties is separated from the cultured liquid of CHO cell, etc. (1) Actions, selectively transfer sulfate group from a sulfate group donor to the 6-site of the N-sulfoglucosamine residue of a heparan sulfate or a heparin having a sulfate group at the 2-site and a hydroxyl group at the 6-site of a glucosamine residue; (2) substrate specificity, transfers sulfate group to a heparan sulfate or a heparin having a sulfate group at the 2-site and a hydroxyl group at the 6-site of glucosamine residue and does not transfer sulfate group to chondroitin and chondroitin-4-sulfate; (3) optimum pH, pH 6-7; (4) optimum ionic strength, 0.1-0.3M (sodium chloride) and (5) inhibition and activation, inhibited by dithiothreitol and adenosine-3',5'-diphosphate and activated by protamine.</p> |
