PURPOSE: To extract main outer film protein of trachoma chlamydia having a specific average molecular weight as species specific antigen, to utilize it in immune reaction diagnosis, together with an analysis method of the presence of the infection. CONSTITUTION: Main outer protein of trachoma chlamydia has a molecular weight of (38 to 44)×100 daltons, 39.5×1000 daltons on the average, and this group of protein is represented as MP39.5. Basic corpuscle obtained by purifying this is brought into contact with a sarcosil solution and is selectively dissolved to leave an outer film. The residue is dissolved by strong anion cleaning agent which dissolves MP39.5, and MP39.5 is recovered from a cleaning agent solution and is purified to obtain MP39.5 antigen. A specific antibody for MP39.5 antigen can be obtained by a proper innoculation method by using laboratory animal such as mouse, rat, etc. This analysis can be made using a known method by which the existence of bacteria antigen in an animal which has been infected is analyzed.
