<p>PURPOSE:To obtain a new fused cell of a mutant of human B cell lymphoblast and a lymphocyte of the patient of gastric cancer, having a specific character and capable of producing an antigenspecific immunoglobulin e.g. for the prevention, treatment and diagnosis of antigenic diseases. CONSTITUTION:This new human/human hybridoma capable of producing an antigen-specific human lmmunoglobulin is produced by subculturing a human B cell lymphocyte while increasing the 6-thioguanine concentration stepwise, selecting a clone proliferative in a serum-free medium containing 20muM 6- thioguanine, culturing the clone in a medium containing N-methyl-N'-nitro-N- nitrosoguanidine as a mutagenic agent to obtain a mutant resistant to 6- thioguanine and ouabain, exterminating in a medium containing HATO, free from production of IgG and IgM and proliferative in a basal medium containing insulin, transferrin, selenium, ethanolamine, fbeta-mercaptoethanol and serum albumin and fusing the obtained mutant cell with a human lymphocyte of a patient of gastric cancer.</p> |
