PURPOSE:To obtain a new polypeptide capable of exactly detecting an e antigen antibody, useful for diagnosing hepatitis B, etc., having such structure that specific amino acid site(s) of the hepatitis B virus e antigen protein for which the C-gene region of hepatitis B virus genome codes has mutated. CONSTITUTION:This new polypeptide, specifically reactive with an anti-HBVe antigen antibody but inert to an anti-HBVc antigen antibody, thus useful for diagnosing hepatitis B, has such structure that the amino acid 74th site and/or 75th site of the HBVe antigen protein for which the C-gene region of HBV genome codes have been substituted with an amino acid such as serine, lysine, asparagine, cysteine, aspartic acid or glutamic acid for the 74th site and with an amino acid such as leucine, proline, tyrosine, glutamic acid, histidine or alanine for the 75th site. This new polypeptide is obtained by manifesting a recombinant vector subjected to a site-specific mutation of the 74th and/or 75th site(s) of a DNA coding for an HBVe antigen. |
