Improved, selective prodn. of beta-(D)-2',3'-dideoxy inosine comprises: (a) treatment of an alpha- and beta-anomeric mixt. of (D)-2',3'-dideoxyadenosine (DDA) with adenosine deaminase; (b) monitoring conversion of the preferentially deaminated beta-anomer; and (c) recovering (I). Monitoring is to control reaction time and minimise alpha-anomer deamination to maximise prodn. of (I). The adenosine deaminase (ADA) is pref. isolated from calf spleen. It is dissolved in neutral aq. medium for reaction with the DDA, or alternatively, an immobilised prepn. used.
