Analysing fecundity of human or animal sperm comprises: (a) placing drop of soln. contg. 0.5-15 wt. % hyaluronic acid, 20-90% 5 mM calcium chloride, 0.1-5% Triton X, and 0.1-5% K2HPO4/KH2PO4, at pH 6.5-8.0 on microscope slide then drying for 1-2 minutes, (b) adding sperm sample which has been previously washed and prepared; (c) placing cover glass over this and incubating for 30 minutes-48 hours at 25-40 deg. C in atmos. contg. 1-10% CO2; (d) examining slide and concluding that sample is fecund if 10-25% of spermatozoa with head membrane intact have halo of at least 7.5 micrometres. Also claimed is a kit for this method, comprising syringe fitted to flask of test soln., microscope slides and covers, and pipettes for application of sperm.
