PURPOSE:To obtain the subject composition not activated with ADP, containing NADH-dependent glutamic acid dehydrogenase, alpha-ketoglutaric acid (salt), and NADH, not requiring expensive NAD, and good in stability. CONSTITUTION:This ammonia measurement reagent composition not requiring expensive NAD having been necessary as an activating agent, excellent in stability and useful for measuring ammonia, urea form nitrogen, creatinine, etc., is obtained by combining (A) glutamic acid dehydrogenase not activated with ADP, dependent on NADH, stable at approximately 60 deg.C (pH8.3, for 10min), having the optimal pH of 10.5-11.5 (oxidative deamination reaction), stable at a pH of 5-10 (at 25 deg.C for 20hr), having the optimal action temperature of approximately 60 deg.C (oxidative deamination reaction), a specific activity of >=300V/mg, and mol.wts. of approximately 280000 (gel filtration method) and approximately 41000 (SDS-PAGE), (B) alpha-ketoglutaric acid or its salt, and (C) NADH. |
