Determn of DNA after capture by probe immobilised on solid phase

Qualitative and quantitative determn. of DNA on solid phases comprises: (1) covalently bonding a single stranded DNA (capture probe) to a solid phase which has been chemically activated by treatment with an alcoholic soln. of organosilane then heating; (2) hybridising the complementary single-stranded DNA (which is to be detected) to the capture probe, and (3) the target DNA is determined: (a) directly from a marker already present in it; or (b) indirectly using an enzyme conjugate, anti-nucleic acid antibody or second DNA single strand (reporter probe) that hybridise with it. Also new is a device for this process comprising a shaped article having a surface, chemically activated by an adhesion promoter, covalently bound to a capture probe.
