PURPOSE:To efficiently obtain the subject compound for preventing and treating leukemia by culturing a bacterium transduced with a recombinant manifestation vector containing a DNA encoding a polypeptide having human granulocyte colony stimulation factor activity and a specific sequence. CONSTITUTION:Cell strain CHU-2 derived from a human stomatic fundus cancer is cultured and its mRNA is separated by a conventional method and used as a template to synthesize cDNA, which is treated with a restriction enzyme, bonded to a vector and transduced to Escherichia coli to give a cDNA library. Then the cDNA library is screened by using a synthetic oligonucleotide probe, a positive clone is selected, its DNA is recovered and treated with a restriction enzyme to give a DNA encoding a polypeptide having human granulocyte colony stimulation factor activity and an amino acid sequence of the formula ((n) is 0 or 1). Further the DNA is linked to a vector, transduced to a bacterium, which is transformed. The transformant is cultured to give the objective polypeptide having human granulocyte colony stimulation factor activity. |
