PURPOSE:To obtain a new polypeptide, having a specific amino acid sequence, capable of manifesting inhibiting activities against trypsin and utilizable for development of a therapeutic agent for diseases such as pancreatitis, shock, multiple organ failure or disseminated intravascular coagulation syndrome. CONSTITUTION:This new polypeptide is obtained by suspending a human cell T98G in a culture medium containing 10% fetal bovine blood serum, culturing the human cell at 37 deg.C for 2 days under conditions of 5% gaseous carbon dioxide/95% air, then removing the culture solution, adding a new culture medium thereto, further culturing the cell for 2 days, collecting the supernatant, adding ammonium sulfate so as to provide 80% saturation, allowing the prepared mixture to stand at 4 deg.C overnight, then centrifuging the resultant mixture, recovering precipitates, subsequently dissolving the recovered precipitates in a buffer solution, carrying out the gel filtration, collecting a fraction having inhibiting activities against trypsin, passing the obtained fraction through a column in which the trypsin is immobilized and eluting the adsorbed substance with a 0.05% trifluoroacetic acid solution. The obtained polypeptide has an amino acid sequence expressed by the formula and is capable of manifesting the inhibiting activities against the trypsin and useful as a therapeutic agent for diseases in which the trypsin participates. |
