摘要 NEW MATERIAL:A proline acylase of about 560,000mol.wt. (according to the gel filtration), having the following physical and chemical properties: Action: acts on N-acyl-L-proline to form L-proline and an organic acid; optimal pH: pH6.0 in potassium phosphate buffer solution; the optimal temperature: 50 deg.C; heat stability: about 12% residual activity after treatment at 60 deg.C for 10min; pH stability: stable in a range from 6-10 pH; inhibitor: almost completely inhibited by Cu<2+> ion and mercuric p-chlorobenzoate. USE:As it hydrolyzes an N-acyl-DL-proline asymmetrically into N-acyl-D-proline and L-proline, it is useful as a catalyst for preparation of L-proline. PREPARATION:Rhodotorula rubra AHU3243 is cultured in a medium containing 0.1-1.0% of benzyloxycarbonylglycyl-L-proline or acetyl-L-proline at 5-8 pH, 20-40 deg.C for 15-40hr, then the cell bodies are crushed, and the enzyme is isolated from the crushed mixture.
