PURPOSE:To obtain a gene included in a DNA corresponding to a fragment of a specific size obtained by the partial decomposition with restriction enzymes MBoI and Sacl and capable of increasing the titer for producing mycinamicin or other antibiotic substances when introduced into a host. CONSTITUTION:This gene participating in the production of mycinamicin and effective for increasing the production titer of mycinamicin and other antibiotic substances when introduced into a host is produced by inoculating a mycinamicin-producing microorganism belonging to the genus Micromonospora [e.g. Micromonospora griseorubida A11725 (FERM BP-3202)] to a culture medium, culturing at 28 deg.C for 2 days under shaking, collecting the microbial cells by centrifugal separation, extracting whole DNA from the cells by SDS-phenol method, etc., partially breaking the DNA with restriction enzymes MboI and SacI to obtain a DNA fragment containing the gene participating in the production of mycinamicin together with various other DNA fragments, cloning the fragment and separating from the DNA corresponding to the fragment of 1.7kb. |
