Prepn. of dye-bio-molecule conjugates

Prepn. of dye-biomolecule conjugates comprises use of leuco-dyes. The biomolecule contains carboxyl gps. and is a nucleic acid, protein or polysaccharide (or a combination of these). The amt. of carboxyl gps. in the biomolecule may be increased by incorporation of p-L-amino acids e.g. p-L-glutamic acid. The dye contains nucleophilic gps. and in its oxidised form does not have sufficient nucleophilic activity to bind to carboxyl gps. Before, or during, coupling to the carboxyl gps., the dye is converted to its leuco form. The carboxyl gps. are opt. activated, e.g. using carbodiimide cpds. or by the formation of activated esters.
