Urokinase is purified by elution from a precipitate with a dilute aqueous solution of 6,9-diamino-2-ethoxyacridine. Alternatively, the urokinase and impurities may be absorbed on calcium phosphate gel in the presence of 6,9-diamino - 2 - ethoxyacridine and the urokinase alone removed by extracting with a phosphate buffer. In a further alternative a urokinase-containing precipitate is washed with acidulated water at pH 3.2-5.5 and the urokinase then extracted with acidulated water at pH 1.5-3. Further purification is by adsorption on carboxymethyl cellulose followed by elution, dialysing and salting out which produces urokinase in crystalline form.ALSO:Purified crystalline urokinase free from pyrogens, thromboplastic substances and potentially antigenic impurities (see Division C3) may be used to treat intravascular clots in humans without producing allergic reactions.
