<p>PURPOSE:To efficiently carry out the culturing by adding a 5,6- dichloroindoleactic acid derivative to a culture medium in obtaining an adventitious embyro, induced from a carrot callus and having a high normal germination ratio. CONSTITUTION:This adventitious embryo is obtained by culturing an explant of a carrot (a growing point, a seedling hypocotyl, a cotyledon, a root, etc.) or a callus induced from the explant of the carrot in a culture medium containing a 5,6-dichloroindoleacetic acid derivative therein. The adventitious embryo thus obtained is capable of normally germinating at a high frequency. 5,6-Dichloroindoleacetic acid or its salt or ester is especially preferred as the 5,6-dichloroindoleacetic acid derivative. The resultant adventitious embryo is useful as an embryo used in an artificial seed or for producing a healthy cultured seedling. Furthermore, the explant for obtaining the callus is preferably collected from a slice of the seedling hypocotyl germinated under aseptic conditions or a slice prepared by sterilizing a root and preferably punching the slice with a cork borer.</p> |
