PURPOSE:To decolor human serum albumin useful, e.g. for therapy of e.g. hypoalbuminemia of fetal erythroblastosis caused by surgery, shock, burn or edema by treating human serum albumin manifested by a gene manipulation with a reducing agent. CONSTITUTION:A recombinant plasmid integrated with a gene coding human serum albumin is transferred to Pichia pastoris GTS 115 (his 4) to transform it and the resultant transformant is inoculated in a culture medium and subjected to shaking culture at 30 deg.C for 24 hr. The culture solution is subsequently compressed to separate the culture supernatant and the resultant culture supernatant is treated with an ultrafiltration membrane having 300000 differential molecular weight. Further, it is treated with another ultrafiltration membrane having 30000 differential molecular weight and the resultant fraction is treated with a cation-exchanger, a hydrophobic chromatography and an anion-exchanger. This treated fraction is then treated with a reducing agent such as sodium sulfite under a condition of pH 9 at 37 deg.C for 24hr, thus decoloring and purifying the objective human serum albumin useful, e.g. for therapy of, e.g. hypoalbuminemia or fetal erythroblastosis. |
