ANTI-NITROGEN MONOXIDE SYNTHASE MONOCLONAL ANTIBODY, ITS PRODUCTION AND USE

PURPOSE:To obtain the subject new monoclonal antibody showing integrity to an NO synthase, useful for measuring the NO synthase simply and in high sensitivity and purifying the NO synthase, comprising an induction type nitrogen monoxide (NO) synthase as an immunogen. CONSTITUTION:An NO synthase derived from macrophage or a brain such as nerve cell or glia cell is administered to a BALB/c mouse, which is immunized. Three days after the final administration of the antigen, a spleen is extracted to collect a cell to be used for cell fusion. Then the cell is subjected to cell fusion with a myeloma cell of mouse and selectively cultured in a HAT medium to give a hybridoma cell. Then the hybridoma cell is cultured, a clone capable of producing an antigen to be reacted with an NO synthase is selected and the positive clone is cloned. The prepared monocloned hybridoma is cultured to give a monoclonal antibody showing integrity to an NO synthase, useful for measuring the NO synthase simply and in high sensitivity and purifying the NO synthase.
