[Also in Division C3] Nitrogen-containing polysaccharides are extracted from unicellular green algae by successively extracting the algae under the action of heat with (i) a hydrophilic organic solvent or a mixture thereof and water, (ii) water, (iii) an alkaline aqueous solution and (iv) an acidic aqueous solution to produce four separate extracts; concentrating these extracts and removing the hydrophilic solvent and alkaline and acidic substances from their respective extracts; forming a precipitate of impurities by adding ethanol to each extract until the ethanol concentration reaches about 80% and removing the precipitates and added ethanol; and finally forming a white precipitate of the desired polysaccharides by adding absolute ethanol to each extract, and recovering the precipitates. The hydrophilic solvent may be methanol, ethanol or acetone. NaOH and H 2 SO 4 respectively are specified as the alkali and the acid. The product has use in stimulating activity of the reticulo-endothelial system. In an example, a rat is injected with a polysaccharide as above, and the increased removal rate of carbon particles previously injected into the bloodstream is noted. |
