Isolation and cloning of cDNA encoding RNA binding protein (RBP)

A method for isolating and cloning a cDNA of an RNA binding protein (RBP) comprises the prodn. of a cDNA gene library from a cell line potentially contg. RBP, where: (i) the gene library linked to expression vectors is expressed in host cells already contg. a further expression vector with a marker gene contg. at its 5'-non-translated region the binding site of the RBP and (ii) detection of the presence of cDNA by reduced expression of the marker gene. Pref., the vectors for the cDNA contain a regulatable promoter and the marker gene pref. is the lacZ-gene.
