PURPOSE:To provide the novel enzyme originated from a strain of the genus Trichoderma, catalyzing a reaction for decomposing ascorbic acid into dehydroascorbic acid and water, not acting on D-araboascorbic acid, not inhib ited with NaN3, and useful for clinical examinations, etc. CONSTITUTION:A method for producing a novel ascorbic acid oxidase comprises inoculating a strain [e.g. Trichoderma lignorum APC-9314 (FERM-P-13972)] belonging to the genus Trichoderma on a medium, aerobically culturing the strain at 30 deg.C for 6 days with stirring, filtering the obtained culture product, subjecting the filtrate to an ultrafiltration, adding cold ethanol to the concentrated solution, stirring the mixture in a refrigerator over a night, centrifuging the produced precipitates, dissolving the obtained precipitates in a buffer solution, and subsequently purifying the solution through an anion exchange column. The novel ascorbic acid oxidase acts on ascorbic acid to catalyze its decomposition reaction into dehydroascorbic acid and water, does not act on D- araboascorbic acid, has the optimal pH of approximately 5, has the optimal temperature of approximately 40 deg.C, and is not inhibited with sodium azide. |
