<p>PURPOSE:To obtain a support for peptide synthesis which is made of a biodegradable polymer and can prepare biologically active peptides, protein fragment peptides through its covalent bonds, thus can be used as a vaccine or therapeutic agent of low toxicity in the form that these peptides are bonded to the support. CONSTITUTION:Poly(lactic acid), a biodegradable polymer, is used as a support for peptide synthesis. The peptide synthesizer is filled with grains of poly(lactic acid) and the terminal carboxyl groups are reduced with lithium aluminum hydride into hydroxyl group, and an alpha-amino group-protected amino acid is bonded with the hydroxyl group in the presence of a condensation agent such as diisopropylcarbodiimide. Then, the amino-protecting group is eliminated and amino acids are allowed to be bonded to the remaining amino group in order according to the sequence of the objective peptide, then the amino- protecting groups are eliminated to give a biologically active peptide or protein fragment peptide bonded to the support through the covalent bond. The support bearing peptide through covalent bonds can be directly given to living bodies as a vaccine or a therapeutic agent of low toxicity.</p> |
