In a process for the enrichment and purification of L-asparaginase obtained from Escherichia coli by digestion of the cell suspension and subsequent manganese precipitation, the supernatant or filtrate from the manganese precipitation is treated with a gel and the gel is separated and eluted at a pH between 8À5 and 10. Suitable gels are those of calcium phosphate, aluminium hydroxide, silicic acid and stannic acid. Further conventional purification steps may include alcohol fractionation, protamine precipitation and chromatography on DEAE-groupcontaining cross-linked dextran. |
