摘要 PURPOSE:To obtain a new L-fucose dehydrogenase applicable to the determination of L-fucose or clinical tests and fundamental research. CONSTITUTION:This L-fucose dehydrogenase is obtained by culturing a microorganism, having the L-fucose dehydrogenase productivity and belonging to the genus Agrobacterium and has the following physico-chemical properties: (A) substrate specificity: acting on L-fucose, L-galactose and D-arabinose; (B) molecular weight: 41000+ or -5000 daltons (measured by an electrophoresis of polyacrylamide gel electrophoresis containing SDS); (C) isoelectric point: pI 14.4+ or -0.5; (D) optimum pH: 7.5-10.0 (30 deg.C); (E) pH stability: 7-11 (at 4 deg.C for 24 hr); (F) optimum temperature: about 50 deg.C) (pH7.5) and (G) temperature stability: stable to about 40 deg.C (pH7.5 for 10min). The L-fucose dehydrogenase is prepared by culturing Agrobacterium tumefaciens K-28 (FERM BP-3417) without requiring an inducer.
