PURPOSE:To provide an imprinting U2afbp-rs gene originated from mouse genom, expressing exclusively the father-side chromosome, preventing the generation of diploid embryo, relating its abnormality to genetic diseases and cancers and forming a genetic product recognizing the acceptor site of intron. CONSTITUTION:The objective imprinting U2afbp-rs gene which is a straight- chain double-stranded cDNA originated from mouse genom and having a base number of 2,831 can be produced by synthesizing a cDNA using a mouse liver RNA as a template, producing a lambdagt11cDNA library by conventional method, screening a white plaque, separating three clones, recovering DNA from the clones, treating with restriction enzymes, fractionating by electrophoresis in polyacrylamide gel, eluting the fragment from the gel, subcloning in a plasmid vector, amplifying with a primer by PCR reaction and screening the amplified product by a restriction landmark genom scanning method using a methylation sensitive restriction enzyme as a landmark. |
