The new antibiotic lipoxamycin, produced by aerobically cultivating Streptomyces - virginiaevar lipoxae (NRRL. 3630), is characterized as follows: analysis: C=61À26, H=9À74, N=7À52, O=21À48; empirical formula: C 19 H 36 N 2 O 5 ; melting point: 68-70‹ C. (d); optical rotation: [α]25 D =-2 (C= 0À877 in MeOH); I.R. absorption (in mull): 2930 (mull), 2900 (mull), 2840 (mull)' 1700, 1650, 1640, 1567, 1460 (mull), 1410, 1375 (mull), 1365, 1200 and 1090cm.-1. The lipoxamycin as prepared and defined above is a mixture of mainly lipoxamycin itself together with other undefined antibiotics. It may be purified as its sulphate which has the following properties: analysis: C=54À13, H=8À85, N=6À64, S = 3À80, O = remainder; Melting point, 153À3‹ C. (d); optical rotation: [α]#25 D = - 10 degrees (C= 0À696 in dimethylsulphoxide); U.V. absorption (in MeOH): # max. =208 mÁ, #=5450 and shoulder at 275 mÁ, #=63; I.R. spectrum as in Fig. 1; formula: C 19 H 36 N 2 O 5 .Y 2 H 2 SO 4 . Lipoxa mycin is amphoteric and forms acid addition salts (e.g. toluene sulphonate, bromobenzenesulphonate, acetate oxalate hydrochloride and iodate) and base addition salts. The antibiotic is obtained from the filtered fermentation beer by extraction with water immiscible solvent, e.g. methylen chloride at alkaline pH, precipitation from the extract as an acid addition salt by adding an appropriate acid, e.g. H 2 SO 4 , recrystallization of the salt, e.g. from methanol and finally purified, after conversion to the free base form by counter-current distribution, e.g. between ethylacetate, ethylene glycol monomethyl ether water (2 : 1 : 1), and precipitated as the salt by adding an acid or isolated by evaporating the solvent. Alternatively the lipoxamycin may be adsorbed on an anionic, cationic or other resin or adsorbent and subsequently eluted with alkali, acid or solvent. Pharmaceutical compositions haying antibacterial and antifungal activity comprise lipoxamycin or lipoxamycin mixture as defined above, or an acid-addition salt thereof together with an appropriate diluent or carrier.
