<p>PURPOSE:To obtain the subject cell the strain BGM-BB5 that is inducted from BGM cell originated from kidney cell of Cercopithecus aethiops capable of stably producing vaccine virus at a low cost by cell free culture and preventing nonspecific reaction at the diagnosis of mycoplasmatic disease. CONSTITUTION:BGM cell originated from kidney established cell of Cercopithecus aethiops is subcultured every week starting from 10% fetal calf serum(FCS) to 4% by reducing the FCS concentration at 2% point every subculture to acclimate. Then, from 4% FCS to 1% the cell is subcultured every week by reducing FCS at 1% point, while changing the medium composition and adding insulin, transferrin, ethanol amine and sodium selenite as growth factors, as the FCS concentration decreases. Subsequently, the subculture is continued until FCS free is attained by reducing FCS at 0.3% point followed by cloning to obtain the objective new strain BGM-BB5 (FERM P-13626), inducted from a BGM cell, capable of serum free culturing.</p> |
