PREPARATION OF STRAIN PRODUCING HUMAN MONOCLONAL ANTIBODY

PURPOSE:To obtain a human monoclonal antibody-producing strain capable of producing a human monoclonal antibody continuously usable as a treating agent by transplanting human lymphocyte to a severe combined immunodeficiency mouse, immunizing the mouse with an antigen and establishing the obtained sensitized lymphocyte. CONSTITUTION:Human peripheral blood lymphocyte is separated from a human leukocyte fraction and transplanted to the abdominal cavity of a severe combined immunodeficiency mouse. An antigen (e.g. CD2 antigen) is applied to the abdominal cavity of the severe combined immunodeficiency mouse transplanted with human lymphocyte to effect the immunization of the mouse. When the antibody value is increased, the spleen and the mesenteric lymph node are extracted from the mouse and separated into single cells using a polished slide glass to separate the sensitized lymphocyte. The obtained sensitized lymphocyte is mixed with a human myeloma cell strain to effect the cell fusion, the fused cell is cultured in HAT medium and the obtained hybridoma is cloned and established to obtain a human monoclonal antibody producing strain in high yield.
