PURPOSE:To obtain the novel strain bacterium capable of producing an enzyme capable of synthesizing steroid, vitamin D3, etc., by binding a rat liver 450C25 gene to the downstream of a promoter of a yeast alcohol dehydrogenase gene and expressing this enzyme in the yeast. CONSTITUTION:A plasmid containing a rat liver 450C25 gene is digested with a restriction enzyme to isolate the rat liver 450C25 gene and the gene is bound to the downstream of the promoter of a yeast alcohol dehydrogenase (ADH) gene to prepare a recombinant plasmid. A recombinant plasmid containing this rat liver P450C25 gene is transformed by a protoplast method, etc., using a yeast such as Saccharomyces cerevisiae AH 22 strain (ATCC38626), etc., as a host to provide the objective novel rat liver P450C25-producing strain capable of mass-producing a rat liver P450C25 enzyme capable of synthesizing steroids, active vitamin D3, etc., useful as a medicine. |
