PURPOSE: To convert a certain subtype of a type A cell of an erythrocyte to a type O cell and to use the obtained cell as type O cell in transfusional therapy. CONSTITUTION: The objective type O erythrocyte is produced as follows: (i) Aint or A2 erythrocytes are equilibrated to a pH of 5.6-5.8, (ii) then the equilibrated Aint or A2 erythrocytes are brought into contact with α-N- acetylgalactoseaminidase for a period sufficient to convert the A antigens of the Aint or A2 erythrocytes to the H antigens, and thus the type O erythrocytes are produced, (iii) the enzyme is removed from the obtained type O erythrocytes, and (iv) the O erythrocytes are re-equilibrated to a pH of 7.2-7.4. The obtained type O erythrocytes contain (a) at least 2,000,000 of terminal α-fucose parts, (b) substantially no terminal α-N-acetylgalactoseamine part and (c) adenosine-5'- triphosphate (ATP) on the level of 60-90% of the level of ATP in the naturally occurring A2 B erythrocyte. |
