<p>This invention provides novel methods and compositions for use in the efficient and large-scale production of recombinant adeno-associated virus (AAV). Described herein are new producer cell lines, recombinant adenovirus or herpes virus vectors and AAV constructs. Also disclosed are particularly advantageous methods of using such materials to produce recombinant AAV virions using only the efficient process of viral infection, without requiring transfection steps. The AAV produced may be used in a variety of embodiments including, for example, for transferring exogenous genes into human cell lines and for use in human gene therapy regimens.</p> |
