PURPOSE:To obtain the subject protein useful for understanding the control of gene manifestation of a fat-soluble physiologically active substance and for development of a new medicine by cloning cDNA intranuclear receptor from an HOS cell of human osteoblast-like osteosarcoma cells. CONSTITUTION:Cultured HOS cell of human osteoblast-like osteosarcoma cells is homogenized in an aqueous solution of guanidium thiocyanate, etc. According to the method invented by Chirgwin et. al., a cDNA fragment is obtained through, e.g. separation of the whole RNAs by the cesium chloride equilibrium density gradient centrifugation method, the sucrose density gradient centrifugation method, etc., and the obtained cDNA segments are cloned, thus affording the objective polypeptide-like protein represented by an amino acid sequence compound of 579 amino acids. |
