NEW MATERIAL:A biphenyl metabolism gene having function of converting biphenyl derived from Pseudomonas putida or a derivative thereof to benzoic acid or a derivative thereof. USE:Producing an agent for decomposing and removing substances of environmental pollution, such as PCB. PREPARATION:For example, Pseudomonas putida KF 715 strain (FERM P-10697) assimilating biphenyl is cultured in a medium, the bacteria are collected, suspended in a buffer solution, the bacteria are subjected to bacteriolysis to extract chromosomal DNA. Then the chromosomal DNA is treated with a restriction enzyme, bonded to a vector and transduced to Escherichia coli, which is cultured to give a culture mixture. A colony which forms 2-hydroxy-6-oxo-6- phenyl-hexa-2,4-dienoic acid and yellows by adding 2,3-dihydroxyphenyl solution to the culture mixture is selected, a plasmid is extracted from the colony and treated with a restriction enzyme to give new gene bphABCD. |
