A method of obtaining a nucleic acid from a sample of blood comprises the steps of: treating the blood in vitro with a high molecular weight carbohydrate having negative charges along the carbohydrate backbone, allowing the red blood cells to sediment, and obtaining a sample of the supernatant liquid, lysing cells in said sample of the supernatant with a lysis mixture containing a surface active agent and a protein digesting enzyme.
