<p>PURPOSE:To obtain a new tetrahydrofurfuryl alcohol derivative having a nucleic acid base being applicable over a wide range of fields such as nucleic acid-based organic chemistry and medicines and molecular biology. CONSTITUTION:A compound of the formula I (B is purine or pyrimidine base; R<1> is H, OH-protecting group, monophosphate residue, diphosphate residue, triphosphate residue; R<2> is H, alkyl, halogenated alkyl; R<3> is H, alkyl; either of X or Y is O and the other is CH2), for example, the compound of the formula II (Ad adenine residue; TBDMS is t-butyldimethylsilyl). This compound is obtained by nucleophilic substitution reaction of nucleic acid base and a compound of the formula V which is obtained by cyclization reaction accompanied by elimination of R<7> I between an olefinic derivative of formula the III or IV (R<4> is R<1>; R<5> is R<2>; R<6> is R<3>; R<7> is H, alkyl, alkylsilyl) and iodine molecule. This compound can be utilized as a chain terminator needed in the Sanger method for analysis of gene, identification and detection of a variety of microorganisms.</p> |
