PURPOSE:To obtain 3-cyano-6-chloropyridine which is a synthetic intermediate for medicines, agricultural chemicals, dyes, etc., by hydroxylating 3-cyanopyridine by microbial reaction and chlorinating the resultant 3-cyano-6-hydroxypyridine by chemical synthetic reaction. CONSTITUTION:3-Cyanopyridine is hydroxylated with a microbial cell of Comamonas acidovorans (NCIMB 9289), etc., or its microbial cell-treated material. The resultant 3-cyano-6-hydroxypyridine is chlorinated with phosphorus oxychloride, etc., to afford the objective 3-cyano-6-chloropyridine. 3-cyano-6- chloropyridine is reduced to give the objective 3-aminomethyl-6-chloropyridine, which is further alkylated with 1-6C alkyl group to provide the objective 3-(alkylamino)methyl-6-chloropyridine. 3-Cyano-6-chloropyridine is reduced in an acidic aqueous solution to provide the objective 3-hydroxymethyl-6- chloropyridine. |
