PLANT TISSUE CULTURE METHOD FOR TAXOL PRODUCTION

A method for producing a Taxus species hairy root culture, by isolating an embryo from a Taxus species seed; germinating the embryo to provide a germling; inoculating the germling or a piece thereof with a transforming strain of Agrobacterium rhizogenes; transferring the inoculated germling to a transformation culture medium e.g., containing either 1-napthaleneacetic acid and Nicotiana species feeder cells, or acetosyringone; transferring the germling from the transformation culture medium, after a period of time sufficient for the Agrobacterium rhizogenes to transform the germling, to an antibiotic culture medium comprising an amount of an antibiotic sufficient to kill the Agrobacterium rhizogenes; selecting a segment of hairy root from the antibiotic culture medium after a period of time sufficient to substantially kill the Agrobacterium rhizogenes; and transferring the segment of hairy root to a liquid or a solid maintenance medium.
