PURPOSE:To obtain a new IgE-binding peptide, having a specific amino acid sequence and the ability to bind to a mite allergen-specific IgE and useful as a desensitizing therapeutic agent, etc., for allergic diseases without any fear of anaphylactic shock. CONSTITUTION:This IgE-binding peptide is obtained by extracting the whole RNA from a live mite insect body according to a conventional method, separating an mRNA from the whole RNA extract solution utilizing the affinity for a poly(A), using this mRNA as a template, synthesizing a cDNA, then integrating the cDNA in which an EcoRI linker is ligated to the EcoRI site of a lambda-gt11 phage, preparing a cDNA library, infecting Escherichia coli therewith, converting a plaque manifesting the positiveness to a western blotting method into a positive clone, recovering the resultant DNA in the positive clone, treating the recovered DNA with a restriction enzyme, integrating the treated DNA into an expression vector, transducing the obtained expression vector into Escherichia coli, transforming the Escherichia coli and culturing the prepared transformant. The IgE-binding peptide has formula I (A to E are amino acid residue) or formula II and the ability to bind to a mite allergen-specific IgE. |
