PURPOSE:To produce the subject new allergen having a specific amino acid sequence, free from an impurity inducing anaphylactic shock, safe, effective and useful as a treating agent for mite allergen diseases, etc., by the expression of a gene derived from a mite body. CONSTITUTION:An mRNA is prepare by extracting a total RNA by a conventional method from living mite bodies obtained by culturing Dermatophagoides farinae, then treating the total RNA with poly (A) to separate an mRNA fraction and synthesizing a cDNA by using the mRNA as a template. A cDNA library is prepared from the cDNA and a positive clone containing the DNA coding for a mite ellergen protein is selected from the library by an immunological screening. The DNA is collected from the positive clone, combined with an expression vector after treated with a restriction enzyme and transduced into a host e.g. Escherichia coli to lead a genetic transformation. The objective recombinant mite allergen containing the amino acid sequence of formula and having molecular weight of about 16000 (SDS-PAGE) is produced by culturing the transformant. |
