<p>PURPOSE:To obtain a transgenic animal, having a modified xeroderma pigmentosa(XP) gene of a homozygote modified so as not to make the xeroderma pigmentosa function and useful as a carcinogenic model reflecting a human disease caused by a genetic abnormality. CONSTITUTION:A targeting vector capable of specifically destroying a xeroderma pigmentosa(XP) gene is transduced into an embryonic stem(ES) cell derived from a murine blastocyst to isolate the ES cell having a modified gene in which the XP gene is destroyed by the homologous recombination. The isolated ES cell is then injected into an animal embryo and transplanted to the uterus of an allomother to afford a chimera animal of a genital series. The resultant chimera animal is mated with a normal animal to provide heterozygotes, which are mutually mated to prepare a homozygote. Thereby, a transgenic nonhuman animal is prepared. In the process, the injection of the ES cell into the animal embryo is carried out without removing the zona pellucida in the embryo of the 8-cell phase to provide the objective transgenic animal useful as a carciriogenic model animal reflecting a human disease caused by a genetic abnormality.</p> |
