PURPOSE:To efficiently obtain beta,14alpha-dihydroxy-4-androstene-3,7-dione having action capable of inhibiting proliferation of a human breast cancer cell by culturing a microorganism which belongs to the genus Myrothecium and capable of hydroxylating 4-androstene-3,17dione in a substrate-containing medium and harvesting the product from the cultured mixture. CONSTITUTION:A microorganism which belongs to the genus Myrothecium and having ability capable of producing 6beta,14alpha-dihydroxy-4-androstene-3,17-dione by hydroxylaitng 4-androstene-3,17-dione [e.g. Myrothecium sp. NK-928521 strain (FREM P-13234)] is inoculated into a medium containing 4-androstene-3,17-dione and subjected to submerged culture at 27 deg.C for 3 day. The resultant culture mixture is diluted with water and 3.0% filtration auxiliary is added thereto and the mixture is filtered by Nutsche funnel in which diatomaceous earth is laid and the filtrate is passed through an adsorptive resin column and the adsorbed material is eluted with 60% methanol and the elute is concentrated under reduced pressure and the deposited crystal is purified by recrystallization to efficiently provide the objective androstene derivative. |
