PURPOSE:To obtain a new gene useful for the screening of water diuretic agent and the production of a highly water-permeable artificial membrane and liposome integrated with WCH-1 protein. CONSTITUTION:A vasopressin-controlling channel (WCH-1) cDNA specific to mRNA capable of expressing a water channel locally existing at renal collecting tubule part and complementary to the mRNA. The cDNA is expressed e.g. by the base sequence of formula. It can be prepared by preparing a single stranded cDNA from mRNA of renal medulla of mammals, subjecting to PCR using 5'(T/C)T(T/C/A/G)AA(T/C)CC(T/C/A/G)GC(T/C/A/G)GT(T/C/A/G)AC-3' and 5'-AA(T/C/A/G)(G/C)(T/A)(T/C/A/G)C(G/T)(T/C/A/G)GC(T/C/A/G) GC(A/G) TT-3' as a degenerate primer and screening the renal cDNA library of mammal using the PCR product as a probe. |
