The invention comprises a lytic agent and method of performing a differential cell subpopulation count, of use for example in flow cytometry. The lytic agent comprises an aromatic oxyethanol, an organic buffer with a pK at or near 8.5, and a nonionic detergent. Preferably, the aromatic oxyethanol is 2-phenoxyethanol. The method and lytic agent allow the five part differential determination of leukocyte cell subpopulations from a whole blood sample using flow cytometry methods. |
