<p>PURPOSE:To easily isolate subprotoplast from the pollen tube of Eucalyptus plant, where three germ pores exist. CONSTITUTION:After gathering the pollen of Eucalyptus plant, highly active pollen is sorted by Percoll density gradient centrifugation and allowed to germinate, and then treated with an enzyme solution whose osmotic pressure is adjusted to 0.8-1.2M to isolate and purify subprotoplast from the pollen tube.</p> |
